A Reverse Transcriptase-PCR Assay for Detecting Filarial Infective Larvae in Mosquitoes
BACKGROUND Existing molecular assays for filarial parasite DNA in mosquitoes cannot distinguish between infected mosquitoes that contain any stage of the parasite and infective mosquitoes that harbor third stage larvae (L3) capable of establishing new infections in humans. We now report development of a molecular L3-detection assay for Brugia malayi in vectors based on RT-PCR detection of an L3...

ES cells. This phenomenon has been observed in our laboratory in a number of cases. Clones putatively identified as homologous recombinants by Southern analysis can be quickly checked for random insertion of extra copies of the targeting vector by PCR. Given the small difference in size between the two alleles and use of only two primers common to both WT and mutant DNA, the amplification rate ...
متن کاملDetection of Wuchereria bancrofti L3 Larvae in Mosquitoes: A Reverse Transcriptase PCR Assay Evaluating Infection and Infectivity
BACKGROUND Detection of filarial DNA in mosquitoes by PCR cannot differentiate infective mosquitoes from infected mosquitoes. In order to evaluate transmission risk an assay is needed that can specifically detect infective L3 stage parasites. We now report the development of an assay that specifically detects the infective stage of Wuchereria bancrofti in mosquitoes. The assay detects an L3-act...
متن کاملMultiplex Reverse Transcriptase-PCR Assay for Typing and Subtyping of Influenza A (H5 & H9) Virus in Iran
Avian influenza virus (AIV) infection is a major cause of bird or human mortality and morbidity, therefore the rapid identification of the virus is of important clinical and epidemiological implication. Methods: A multiplex Reverse Transcriptase PCR (RT-PCR) was optimized for the detection of influenza A virus and the H5 and H9 subtypes. The influenza type A specific primers were directed to t...
متن کاملReverse transcriptase-PCR assay for detection of hog cholera virus.
A reverse transcriptase-PCR strategy was developed for the detection of hog cholera virus. Hog cholera virus template was amplified from tissue culture fluids and from tissues and blood of infected pigs, but not from samples containing other pestiviruses. Restriction endonuclease analysis identified samples as historic or recent isolates.
